Pregnancy and postpartum period are associated with demanding physical and psychological changes that often lead to the development of psychological disorders. Depression is diagnosed in more than one in six women after childbirth. However, the prevalence of postpartum depression can be much higher because many cases are undiagnosed. In the case of severe depression, the patient is switched to pharmacological treatment, with sertraline being the most commonly used for this diagnosis. A new drug used in the treatment of postpartum depression is brexanolone, which was registered by FDA in 2019. The advantage over conventional therapy is its rapid onset of action. The structure represents the neuroactive steroid – allopregnanolone, which acts as an agonist on the δ-subunit of the GABA receptor and improves the symptoms of postpartum depression. In addition to the registered brexanolone, another steroidal drug, zuranolone, is available in the third phase of the clinical trial. The steroid structure was chemically altered to improve bioavailability and create an oral dosage form. Another synthetic analogue of neuroactive allopregnanolone, known as ganaxolone, did not show a significant reduction in depressive symptoms in the second phase of the clinical trial compared to placebo. Nevertheless, it has great therapeutic potential in the treatment of various types of epilepsy.

The research was performed on 80 male rats of the Wistar line. Animals of two age groups were used: adults (10–12 months) and old (22–25 months). The obtained data show that the development of immobilization stress in adult and aged rats are accompanied by the formation of a characteristic complex of changes in the phospholipid composition of the myocardium. Intraperitoneal injection of the chokeberry extract (Aronia melanocarpa) at a dose of 0.2 g/kg 60 minutes before the immobilization has limited stress modulation of myocardial phospholipid composition in aged animals. Thus, the extract of Aronia melanocarpa increases the myocardial resistance to the injury effect of stress.

The article presents the results of a preclinical study of ramipril and candesartan in an experimental group of hypertensive rats of different sexes. Antihypertensive therapy was performed for 21 days. The drugs were administered daily in moderate therapeutic doses calculated for rats using the coefficient of species sensitivity. It was found that the course of experimental hypertension has gender differences, and in males, according to blood pressure, the level of NO metabolites is more pronounced. The use of ramipril from the group of ACE inhibitors and candesartan from the ARBs group in experimental hypertension in rats has gender differences. Ramipril is likely to be more effective in normalizing blood pressure and endothelial function in males than females. The use of candesartan did not show significant gender differences, but there was a tendency for females to be slightly more effective than males. Established gender differences in hypertension pharmacotherapy should be considered to optimize treatment.

Metronomic therapy is a therapeutic method in selected oncological diseases, using long-term administration of low doses of drugs with direct or indirect antitumor effect. In addition, to direct cytotoxic eradication of tumor cells, metronomic therapy can very strongly affect the tumor microenvironment; it also has an immunomodulatory and antiangiogenic effect. Its minimal toxic profile allows for use in patients with severe organ dysfunctions and directly impacts the quality of life and social inclusion of oncological patients.

This article presents the results of the study of the antimicrobial and antifungal properties among 1,2,4-triazole derivatives synthesized at the Department of Physical and Colloidal Chemistry of the Zaporizhzhia State Medical University. Previous studies have established the antimicrobial and antifungal activity of 1,2,4-triazole derivatives. Therefore, it was reasonable to investigate highly effective substances with antimicrobial and antifungal properties among synthesized compounds. In the first stage of our research, acute toxicity prediction was performed. The antimicrobial and antifungal properties were carried out by the method of “serial dilutions” on a liquid nutrient. Forty-seven compounds of the different classes were studied for these types of activities. According to our research, derivatives of 3-amino-1,2,4-triazole showed better performance than 3-thio-1,2.4-triazoles for Staphylococcus aureus and Candida albicans. 5-(1Н-tetrazole-1-іl)methyl-4Н- -1,2,4-triazole-3-yl-1-(5-nitrofuran-2-yl)methanimin (11.6) was showed the greatest antimicrobial and antifungal activity. Deeper research for compound 11.6 was done by diffusion in agar (method of “wells”). Studies have shown that molecule 11.6 showed antimicrobial and antifungal action to the studied test strains at a concentration of 2 μg/ml. Hence, this compound can be developed as a helpful therapeutic agent after establishing its safety pharmacology and toxicity.

This study aimed to determine the amount of energy to damage the red galangal rhizome sliced cell tissue and the amount and quality of the essential oil obtained by steam-water distillation. This study was a randomized block design, with pulsed electric field (PEF) voltage treatment starting at 1000, 2000, 3000, 4000, and 5000 V and without PEF and repeated three times. The results showed that the voltage between 3000 and 4000 V (E = 120–160 V/cm) or equivalent to 271.5–365.0 kJ/cm3 had damaged the red galangal rhizome slice cell tissue. The increase in yield due to PEF ranged from 13% to 73%, and there was no change in the value of the refractive index and specific gravity, but there was a change in the chemical composition of the essential oil constituents. The benefits of research with PEF treatment are the increase in yield and shorter distillation time.

Metabolic syndrome is diagnosed mainly in people of economically developed parts of the world and it affects 20–25% of the adult population worldwide. Nowadays, it is also more frequently diagnosed in children and adolescents. In addition to standard treatment that often involves polypharmacotherapy, and thus increases risk of side effects caused by drugdrug interactions, it is appropriate to look for alternative tools to support the treatment of metabolic syndrome components. Natural polyphenolic compounds, usually present in the so-called functional foods, are suitable candidates for that matter, due to the bioactivity and beneficial effects on the human body. Quercetin, troxerutin, diosmin, hesperidin or silybin are among the currently studied and used natural polyphenolic compounds with a positive effect on aspects of the metabolic syndrome. In addition to their antioxidant and anti-inflammatory effects, these compounds have other positive properties that very often outweigh their side effects whilst their usage in the pharmacotherapy.

Inhaled medications are considered a preferred dosage form of medicine to treat respiratory diseases, such as bronchial asthma and chronic obstructive pulmonary disease (COPD), when bronchodilators or, more precisely, controlled drugs for treatment are used. The main objective of our research was to find out community pharmacists’ general knowledge of particular inhalation systems (IS) dispensed in pharmacies. All data necessary for the evaluation have been obtained by an anonymous survey questionnaire carried out between September 2020 and January 2021, which was done both in printed form and online. The questionnaire was self-construction, anonymous, consisting of sociodemographic data (age, gender, residence address – region) and 22 research questions. Both the administration of bronchodilators and inhaled corticosteroids were monitored in the survey. A total of 115 respondents participated in the survey, 86 females (75%) and 29 males (25%). Most of the respondents, 45 (39%), were aged between 23 and 29. The data show that 62 respondents (53.91%) encounter inhaler dispensing several times a day, with the prevalence of aerosol inhalers (MDI), which was confirmed by 78 pharmacists (67.83%). Most of the time, pharmacists meet patients with bronchial asthma, 80 (69.57%), and bronchodilators are prescribed for therapy mainly, 74 (64.35%). In total, 60 respondents (52.17%) provide patients with information on proper administration of inhaled medicines at their request, and only 46 of the respondents (40%) do it without being asked. The survey outcomes show that community pharmacists have minimum knowledge of nebulizers and also of the use of inhalator attachments. The vast majority of participants, 90 (78.26%), say that since they graduated, they haven’t attended any education courses aimed at proper administration of inhaled medications as part of their constant education. Up to 95 respondents (82.61%) are interested in IS mock-ups in a pharmacy, 103 pharmacists (89.97%) in information materials for their self-study, and 110 respondents (95.65%) in IS leaflets and brochures for patients. A positive finding of the survey is that the majority of the respondents, 101 (87.83%), are interested in further educating themselves to get knowledge of this matter in the future. Our survey outcomes show that our respondents’ knowledge of inhalation systems (IS) is sufficient. Most of the pharmacists who participated in the survey can provide patients with basic information regarding the proper administration of inhaled medicines.

A variety of supplement-drug interactions has been identified, and therefore health scientists should be aware of the proper usage of nutritional supplements. The main aim of this study was to assess beliefs and knowledge related to nutritional supplements among pharmacy students. A cross-sectional study was conducted in order to assess beliefs and knowledge related to nutritional supplements among pharmacy students. A literature review on nutritional supplements was conducted in order to develop a 24-item questionnaire, and expert indications established its face and content validity. Τhe sample was comprised of 142 pharmacy students in Cyprus, whose results are presented. The majority of the participants was receiving nutritional supplements (66.9%) and believed that nutritional supplements may interact and may have toxic effects. Those in the higher years of study scored higher in knowledge than those in the lower ones. The mean scores (mean ± SD) on the knowledge about nutritional supplements by the years of study were 9.14 ± 2.67, 8.71 ± 2.92, 9.91 ± 2.08, 11.16 ± 3.00, and 15.76 ± 2.67 and compared using one-way ANOVA followed by Bonferroni correction. The level of statistical significance was set at p < 0.05. The study showed that pharmacy students are adequately informed about nutritional supplements and that this subject is essential for the curriculum of pharmaceutical studies.

The aim of the present study is the development and validation of a simple method based on capillary zone electrophoresis coupled with UV detection for simultaneous determination of tramadol and paracetamol in pharmaceutical and biological samples. The background electrolyte was composed of 50 mM ammonium carbonate, which is a type of a non-conventional electrolyte system. The developed method was characterized by suitable validation parameters, such as linearity (coefficient of determination r2 ≥ 0,995), selectivity or the limit of detection at the level of 0.25 – 0.5 μg/ml. Acceptable values of accuracy and precision were obtained, which were in good agreement with the recommended validation guidelines for analysis of pharmaceutical and biological samples. Detection was performed at a wavelength of 200 nm. The developed method was successfully applied to determine tramadol and paracetamol in various dosage forms and in urine biological samples. Achieved results indicate a potential of the method to be integrated in the common quality control processes of drugs and/or in bioanalysis.

The prevalence of epilepsy in the world population together with a high percentage of patients resistant to existing antiepileptic drugs (AEDs) stimulates the constant search for new approaches to the treatment of the disease. Previously a significant anticonvulsant potential of cardiac glycoside digoxin has been verified by enhancing a weak activity of AEDs in low doses under screening models of seizures induced by pentylenetetrazole and maximal electroshock. The aim of the present study is to investigate the influence of digoxin at a sub-cardiotonic dose on the anticonvulsant activity of valproate, levetiracetam, and topiramate in models of primary generalized seizures with different neurochemical mechanisms. A total of 264 random-bred male albino mice have been used. AEDs were administered 30 min before seizure induction once intragastrically at conditionally effective (ED50) and sub-effective (½ ED50) doses: sodium valproate and topiramate – at doses of 300 and 150 mg/kg; levetiracetam – at doses of 100 and 50 mg/kg. Digoxin was administered once subcutaneously at a dose of 0.8 mg/kg body weight (1/10 LD50) 10–15 min before seizure induction. Picrotoxin (aqueous solution 2.5 mg/kg, subcutaneously), thiosemicarbazide (aqueous solution 25 mg/kg, intraperitoneally), strychnine (aqueous solution 1.2 mg/kg, subcutaneously), camphor (oil solution 1000 mg/kg, intraperitoneally) have been used as convulsive agents for seizure induction. It was found that under the conditions of primary generalized seizures induced by picrotoxin, thiosemicarbazide, strychnine, and camphor, digoxin not only shows its own strong anticonvulsant activity but also significantly enhances the anticonvulsant potential of classical AEDs sodium valproate, levetiracetam, and topiramate. The obtained results substantiate the expediency of further in-depth study of digoxin as an anticonvulsant drug, in particular, the in-depth study of neurochemical mechanisms of its action.

The main component of Ophiocordyceps sinensis and Cordyceps militaris extracts are polysaccharides. These are natural biopolymers that represent a large class of biologically active components. These contribute to their pharmacological activity and effect on health. They contain monosaccharides that include rhamnose, ribose, arabinose, xylose, mannose, glucose, galactose, mannitol, fructose, and sorbose. The exopolysaccharide fraction has a large number of pharmacological effects, the two most important of which are immunomodulatory and antitumour. Among the contained polysaccharides is also mannoglucan, which shows weak cytotoxic activity against the SPC-I1) cancer cell line. More than ten nucleosides and their related compounds, including adenine, adenosine, inosine, cytidine, cytosine, guanine, uridine, thymidine, uracil, hypoxanthine, and guanosine, have been successively isolated from Ophiocordyceps sinensis. It contains many amino acids and polypeptides that are thought to affect the cardiovascular system. They also have a sedative and hypnotic effect, with tryptophan being the most effective component among them. Polysaccharides were extracted from four samples: sample 1 (grown on the substrate Oryza sativa indica, strain Ophiocordyceps sinensis), sample 2 (grown on the substrate Oryza sativa japonica, strain Ophiocordyceps sinensis), sample 3 (grown on the substrate Oryza sativa indica, strain Cordyceps militaris), sample 4 (grown on Oryza sativa japonica substrate, strain Cordyceps militaris). Through NMR spectroscopy and subsequent comparison with the literature, the majority of a chemical compound in deproteinized extracts 1 and 4 was found to be a hydrophilic polyglucan referred to as CBHP2).

Films are thin, flexible, and transparent wound dressings. They can be prepared from both synthetic and natural materials. In practice, synthetic polyurethane dominates, but research is mainly focused on substances of natural origin. An endogenous substance with excellent filmforming properties, which is involved in the wound healing process, is collagen. However, collagen films themselves have weak mechanical properties, which can be improved by, among other things, combining collagen with other materials. Such material could be carboxymethylcellulose, which has been shown to affect wound healing positively. Films consisting only of CMC also have weak mechanical properties, so combining both materials seems to be a suitable solution to the given problems, and a wound dressing with many beneficial properties for wound healing could be created. Therefore, our experiment aimed to prepare composite films for wound therapy consisting of a combination of collagen and CMC. The films were prepared by the solvent evaporation method, and their properties were compared with those formed only by CMC. In both cases, films with suitable organoleptic, physicochemical, and application properties for wound therapy were produced. The composite films showed lower absorption capacity and better mechanical resistance compared to those formed only by CMC. The combination of collagen and CMC in composite films intended for wound therapy has thus resulted in improved properties of the resulting dressing and holds potential for further research.

Antimicrobial properties of the new combined dental gel

Given the role of the microbial factor in the development of infectious-inflammatory processes in the oral mucosa, the research aim was to study the antimicrobial action of a new combined dental gel containing Rotocan® (10%) and triclosan (0.4%) in vitro and in the traumatic stomatitis in albino rats. Rotrin- Denta® exhibited strong antimicrobial activity against etalon strains of gram-positive (S. aureus ATTC 6538, S. pyogenes DICK 1, B. subtilis ATCC 6633) and gramnegative (E. coli ATCC 25922) bacteria, which exceeded the action of the reference drug Camident-Zdorovia® and weak effects on pseudomonads (P. aeruginosa ATCC 27853) and fungi (C. albicans CCV 885-653), which is less to the reference preparation. Rotrin- Denta® reduced microbial insemination and eliminated oral dysbiosis in albino rats with traumatic stomatitis, exceeding the effect of Kamident-Zdorov’ya®. The results open up the prospect of its clinical testing and further implementation in the dentistry practice.

The aim of the study was to point out the contribution of new invasive therapeutic procedures in the treatment of advanced stages of Parkinson’s disease (PD) in comparison with classical oral pharmacotherapy. Data originated from a group of 43 patients with PD, 39% (17) with classic treatment, 23% (10) with intestinal gel of methyl ester levodopa (Duodopa), 19% (8) of patients were using subcutaneous delivery of apomorphine (APO) and the same quantity of patients had undergone deep brain stimulation (DBS). Majority of patients had advanced stages of PD, stage 4, by standards of Hoehn and Yahr scale (Hoehn and Yahr, 1967). Research observed improvement in majority of patients with novel treatments. A positive effect was also noted in the reduced need for oral therapy, where there was a significant decrease in all new therapies. Benefits were observed in the amount of antiparkinsonic drugs taken per os, where we observed reduction in all new therapies. A positive effect of the new therapeutic approaches in reducing “off” periods in patients has also been noted. In the case of Duodopa and DBS, the ”off” period was shortened up to 50% and in the apomorphine pump up to 40%. Patients also reported reduction of some symptoms like rigidity, tremor and bradykinesis while dyskinesis still remains suba challenge. On the basis of the obtained results, it can be concluded that new therapeutic procedures for PCh will make it possible to manage symptoms typical of advanced stages of the disease, which without these procedures would lead to disability, which is the main reason for their indication. However, in early stages, well responding patients or in slow progressing disease oral antiparkinsonics are remaining as golden standard of treatment. This is not just due to good response but also because these classic drug formulations are significantly less expensive. In Slovakia, novel treatments are accessible through healthcare insurance only after secondary revision by insurance company doctors.

The concept of pharmaceutical care (PC) has existed as a professional philosophy for more than 30 years. However, for a long period of time, little had been done for its integration into the regular practice of healthcare provision. The COVID-19 pandemic and the resulting increase in patient influx in the community pharmacies (CP) encouraged the exploration and establishment of new healthcare services provided within the CP. Nevertheless, these services of PC are still novel, and more can be done to expand the community pharmacists’ current role in primary healthcare. This can be achieved by improving and expanding the newly established services, all while incorporating new ones, for the benefit of public health and the reduction of avoidable healthcare expenditures. This article reviews information about the benefits of this service regarding patient health and the reduction of financial expenses pertinent to adverse drug events within the setting of the CP. Adverse drug events account for significant healthcare expenses and patient distress due to relevant symptoms, emergency doctor visits, and increased hospitalization rates. Several studies conducted internationally have investigated the positive impact of PC practiced by community pharmacists. In spite of results sometimes presenting a non-continuous pattern, PC applied under specific conditions has tangible positive outcomes. Congestive heart failure and type 2 diabetes mellitus patients presented fewer hospital admissions, better symptom control, and higher adherence in comparison to control groups, while a study on asthma patients revealed improved inhalation techniques. All intervention groups reported psychological improvement and a better understanding of their treatment. Special reference is made to the importance of this service for patients receiving anti-cancer treatment and how community pharmacists can have a crucial role in designing, monitoring, and re-designing these therapeutic schemes whose complexity and related adverse drug events negatively affect patient adherence. The role of community pharmacists was very important, especially for primary care, for both patients and healthcare systems during the pandemic, and it seems that it will remain decisive in the post-COVID era as well. The increased complexity of therapy and polypharmacy creates the need for organized, active participation of pharmacists in healthcare provision so that they can use their knowledge and skills under continuous cooperation with other healthcare professionals, thus providing coordinated services for the benefit of the patient.
